Tert-butyl 2-formylpyrrolidine-1-carboxylate (tBFP) is a potent inhibitor of tumor necrosis factor alpha (TNF-α) and other factors that are involved in the inflammatory response. TNF-α is an important proinflammatory cytokine that promotes the production of other inflammatory mediators, such as prostaglandins, nitric oxide, and leukotrienes. TNF-α also has been implicated in the pathogenesis of many chronic inflammatory diseases, such as rheumatoid arthritis and Crohn's disease. tBFP binds to the active site of TNF-α and inhibits its binding to receptor complexes. tBFP has a monodentate ligand, which allows it to bind to one side of the protein target molecule with high affinity. This type of binding is thought to be responsible for the potent inhibition of TNF-α activity by tBFP.
