N-Butyl-N-(4-hydroxybutyl)nitrosamine (BBN) is a genotoxic carcinogen that causes urothelial carcinoma in laboratory animals. It is also a potential biomarker for bladder cancer in humans, based on the detection of BBN in maternal blood and urine. The pharmacological mechanism of BBN is not yet clear, but it may be due to its sodium salt or the presence of other genotoxic chemicals. A rat model has been developed for BBN toxicity studies. This drug inhibits protein synthesis by inhibiting RNA polymerase II and III, which are responsible for gene transcription and translation respectively.
