4-Benzyloxy-2-nitrotoluene is a hexamethyldisilazide derivative that can be synthesized by the reaction of 2-chlorobenzaldehyde and 4-hydroxybenzyl chloride. It has been shown to be an effective chiral building block in organic synthesis. This compound undergoes an oxidative cyclization to form a six membered indole ring system, which can then be coupled with methylmaleimide or acetylation to yield the desired product. The synthesis of 4-benzyloxy-2-nitrotoluene is achieved through the use of enantiomerically pure reagents, such as prochiral hexamethyldisilazide and diastereomerically pure methylmaleimides. The final product is obtained after coupling with either a chloride or bisindolylmaleimide, followed by oxidation with arcyriaflavin.
