Bicuculline methiodide is a potassium channel blocker that is used to study the function of 5-HT2 receptors in the central nervous system. It blocks the flow of anions and cations across neuronal cell membranes, which leads to an increase in cytosolic calcium levels and subsequent depolarization. Bicuculline methiodide has been shown to inhibit locomotor activity and decrease blood pressure in vivo models. It also blocks ATP channels and glutamate-activated channels, leading to inhibition of neurons. In vitro experiments have shown that bicuculline methiodide inhibits ion currents through voltage-gated potassium channels with high affinity for these channels. This drug also has been shown to induce seizures in experimental models by inhibiting neurotransmitter release from neurons in the brainstem trigeminal nerve.
