Description:

N-t-Boc-L-proline-N-hydroxysuccinimide ester is a histidine derivative that has been shown to be an effective inhibitor of serine protease. It is used as a substrate analogue for the study of influenza virus and glycosidases. The reaction yield was increased by using trifluoroacetic acid, which is more reactive than hydrochloric acid, in the synthesis of N-t-Boc-L-proline-N-hydroxysuccinimide ester. One drawback to this method is that it requires a solid phase synthesis due to its insolubility in water. This product is also acetylated at the nitrogen atom on the side chain, which may increase its stability and reduce its reactivity with nucleophiles.
