Epacadostat is a tryptophan analog that inhibits the enzyme IDO1 and blocks the synthesis of Tryptophan. It was shown to have inhibitory activities against a wide range of cancer cells, with selectivity for tumor cells over normal cells. Epacadostat has been tested in mouse models and found to be cytotoxic at concentrations as low as 10 μM, which is an effective concentration for inhibiting tumor growth. The IC50 values for epacadostat were determined to be 0.05 μM for leukemia cells and 0.06 μM for colon cancer cells. Epacadostat has shown to be an inhibitor of iddo-1, which is an enzyme that catalyzes the conversion of tryptophan into kynurenine, thereby blocking the synthesis of serotonin and melatonin, which are neurotransmitters that are important in regulating mood and sleep cycles.
