(E)-N-benzyl-2-cyano-3-(3,4-dihydroxyphenyl)acrylamide (TPCA) is a tyrphostin and inhibitor of protein kinases. TPCA targets the proteins that are involved in cancer growth and apoptosis. It has been shown to inhibit the tyrosine kinase activity of c-Src and c-Abl, which are associated with oncogenesis. TPCA inhibits the tyrosine phosphorylation of eosinophil cationic protein (ECP), leading to decreased ECP release from the cell. TPCA also binds to DNA in vitro and has been shown to inhibit nuclear dNA binding activity in tumor tissues. TPCA is a potent inhibitor of DNA polymerase α, which is an enzyme that synthesizes DNA by adding nucleotides onto the 3' hydroxyl group at the end of linear DNA fragments.
