Benzo[a]pyrene-7,8-diol (BPD) is a postulated carcinogen. It is an aromatic hydrocarbon that has been shown to have carcinogenic potential in humans and animals. BPD binds to DNA at the C2 position of guanine, forming DNA adducts with mismatches and oxidative damage. These lesions are repaired by the nucleotide excision repair pathway, which leads to mutations in genes encoding proteins involved in cancer development. BPD has also been shown to cause chromosomal aberrations in human lymphocytes and rat liver cells. The reaction mechanism for BPD is thought to be similar to that of benzo[a]pyrene (BaP), with the difference being that BaP is activated by cytochrome P450 enzymes while BPD does not require this activation step.
BPD may cause cancer by bypassing the detoxification system of cytochrome P450 enzymes.
