Nipecotic acid is a pyrimidine derivative that inhibits the growth of tumor cells. It has shown potent anticancer activity in cell-based assays and in animal models of cancer. Nipecotic acid inhibits the synthesis of DNA, RNA, and proteins, which may be due to its ability to inhibit ribonucleotide reductase and DNA polymerase. Nipecotic acid also blocks the uptake of glucose into mammalian cells. In vivo studies have demonstrated that nipecotic acid inhibits the formation of atherosclerotic lesions in rabbits. Nipecotic acid binds to DNA gyrase and topoisomerase IV enzymes, leading to inhibition of both bacterial and mammalian cells. The stereoisomers are active against tumor cells with different levels of potency: (+) is more active than (-).
