Description:

N-Cyanomethylpyrrolidinium triflate is a 6-phosphate dinucleoside that is insoluble in water. It has been used as an antisense therapeutic to inhibit the synthesis of HIV-1 and herpes simplex virus (HSV) DNA. N-Cyanomethylpyrrolidinium triflate is activated by phosphite and acidic conditions, which are required for its incorporation into DNA and RNA. N-Cyanomethylpyrrolidinium triflate is synthesized using solid-phase chemistry, with phosphorothioates being the nonbridging linkers between nucleobases. The monomers are assembled on the solid support to form polymers. These polymers can be cleaved from the solid support and purified by HPLC or gel electrophoresis.
