It can be synthesized by microwave irradiation of formamide in the presence of chloroacetaldehyde and potassium bromide followed by cyclization with formic acid. This reaction can also be achieved using a formylation reaction with formic acid and n-substituted pyridines. 4-Chloropyrido[3,2-d]pyrimidine is a versatile molecule that has been used for the synthesis of diverse compounds such as 3-chloroformamidinium chloride and bis(4-chloropyrido[3,2-d]pyrimidinium) chlorochromate.
