2’-Chloro-2’-deoxy-5-methyluridine is a prodrug that is converted to 5-fluoro-2'-deoxyuridine monophosphate (5FdUMP) in mammalian cells. This conversion is catalyzed by cytidine deaminase, which converts the 2' position of the sugar moiety to an amino group. In mammalian cells, this conversion occurs at a low rate and requires incubation. The 5FdUMP derivative is then phosphorylated by deoxycytidine kinase to form 5-fluorouracil, which inhibits DNA synthesis and replication. The cytosine residues in DNA are converted to uracil residues, resulting in misreading of the genetic code.
