The synthesis of 4H-cyclopenta[b]thiophen-6(5H)-one is accomplished by bromination of 3-methylcyclohexanone with N-bromosuccinimide. This reaction yields the corresponding bromoketone in high yield. The bromoketone can then be reduced to the desired product, as shown in Scheme 1.

4H-Cyclopenta[b]thiophen-6(5H)-one is an analogue of the natural product 6,7,8,9,10,11-octahydroanthracene. It has been shown that this molecule binds to a number of proteins involved in DNA replication and transcription through hydrogen bonding interactions.
