FK506 is a potent immunosuppressant that is structurally related to cyclosporin A. It binds to the cytosolic protein FKBP-12 and inhibits calcineurin, an enzyme that dephosphorylates the transcription factor NFAT. FK506 has been shown to be effective against depression and potent anticancer agents. FK506 also has anti-inflammatory properties, which may be due to its ability to inhibit prostaglandin synthesis. The compound is biosynthesized by the bacterium Streptomyces tsukubaensis, which produces a number of functional groups including fluorine and hexanoic acid.
