Ceramide is a sphingolipid that is synthesized from the amino acid serine and palmitoyl-CoA. It plays a role in regulating cardiac and cancer cells, as well as insulin-stimulated glucose uptake. Ceramide can also induce apoptotic cell death, which may be due to its ability to inhibit nuclear DNA synthesis and induce cytolysis. Ceramides have been shown to be elevated in fatty acid biopsy samples taken from people with type II diabetes. Genetic ablation of ceramide synthase (CerS) has been shown to protect mice against obesity and insulin resistance.
