6-Chloro-1H-indole-2-carboxylic acid methyl ester is a type 1 allosteric modulator that binds to the CB1 cannabinoid receptor. It was found to be an agonist at the CB1 receptor, with an affinity of 1.3 nM and a functional potency of 3.2 nM. 6CICA has been shown to have type 2 binding properties, which may be due to its amide group and orthosteric binding site. The linker between these two functionalities may also account for this property. 6CICA has been shown to be an allosteric modulator of the CB1 receptor, meaning that it does not activate or inhibit the receptor but rather influences how other substances bind to the receptor.
