5-(Chloromethyl)uracil (CMU) is a drug that is synthesized by the condensation of uridine and dichloroplatinum(II) chloride. It is a cytostatic agent that inhibits the synthesis of DNA and RNA, which are the building blocks of all cells. CMU prevents the incorporation of uridine into DNA, inhibiting DNA synthesis. The cytotoxic effect of CMU is due to its ability to inhibit thymidylate synthase, which leads to decreased production of thymidine triphosphate (a precursor for DNA). This in turn leads to cell death by preventing cell division. 5-(Chloromethyl)uracil also has an anti-cancer activity because it inhibits protein synthesis in tumor cells by blocking the enzyme ribonucleotide reductase.
