4-[(4-Chloro-2-fluorophenyl)amino]-4-oxobutanoic acid (4CFP) is a potent inhibitor of the enzyme acyl coenzyme A:cholesterol acyltransferase (ACAT). ACAT catalyzes the transfer of cholesterol from high density lipoprotein to low density lipoprotein. 4CFP has been shown to reduce cholesterol levels in animal models. The tautomeric forms of 4CFP are also active, and it has been shown that the sulfinyl form is more potent than the carbonyloxy form. The sulfinyl form is a competitive inhibitor of ACAT with respect to both substrates, whereas the carbonyloxy form is a noncompetitive inhibitor with respect to cholesterol.
