Acridone, also known as 3-[[(1,1-dimethylethoxy)carbonyl]amino]-5-fluoro-4-oxopentanoic acid methyl ester, is a drug that has been shown to induce apoptosis in cancer cells. Acridone is an analog of the natural growth factor β1 and binds to the same receptor site on the cell surface. It inhibits protein synthesis by causing caspase activation and mitochondrial membrane potential collapse. Acridone also inhibits the transcriptional regulation of Bcl-2 by binding to c-jun phosphorylation sites and prevents NFκB translocation into the nucleus. Acridone induces apoptosis in carcinoma cell lines and colorectal adenocarcinoma cells by interfering with signal transduction pathways, such as MAPK and AKT kinases.
