4,6-Dimethylnicotinonitrile is an active site inhibitor of acetaldehyde dehydrogenase. This compound binds to the active site of the enzyme and blocks the entrance of substrate. The affinity for this inhibitor is similar to that of anthranilic acid, but its ability to alkylate thiols is lower than that of halides. 4,6-Dimethylnicotinonitrile has been found to inhibit acetaldehyde dehydrogenase in vivo and in silico. The hydroxylamine moiety on the nitrile group provides a possible link between affinity and activity. 4,6-Dimethylnicotinonitrile is an active analogue of phenacyl with a higher potency against acetaldehyde dehydrogenase than phenacyl itself.
