Dimethyl 4-bromopyridine-2,6-dicarboxylate is a chiral ligand that is used in asymmetric catalysis. It has been shown to be an effective catalyst for the alkylation of aromatic halides with thiols. This compound reacts with sodium metal to form the sodium salt, which is soluble in water. Dimethyl 4-bromopyridine-2,6-dicarboxylate can be synthesized by reacting bromine and pyridine in the presence of a catalytic amount of acid. The synthesis proceeds via an intermediate phosphinyloxy intermediate (2) formed from reaction of bromine with dimethylformamide and phosphorus oxychloride. This intermediate reacts with pyridine to give 2,6-dibromopyridine (3). Reaction of 3 with potassium hydrogen sulfate leads to the desired product (1).
