Pyronaridine is a pyrazine compound that has been reinvestigated and optimized for use as an antimalarial drug. It is the active metabolite of 7,10-dichloro-2-methoxybenzo(b)-1,5-naphthyridine (DDMNB). Pyronaridine inhibits the enzyme cyclase and blocks the synthesis of cyclic nucleotides, which are necessary for cell metabolism. The compound also inhibits protein synthesis by inhibiting RNA polymerase II and DNA polymerase III. Pyronaridine is not active against chloroquine-resistant strains of Plasmodium falciparum but does have activity against strains resistant to quinine or mefloquine. The main side effect is gastrointestinal disturbances.
