7-Dimethylaminocoumarin-4-acetic acid (DMACA) is a carbonyl group that inhibits placental alkaline phosphatase. It has been shown to inhibit cellular uptake, efflux pump activity, and biochemical reactions. DMACA is cytotoxic to cells in culture at concentrations of 10 μM or greater and has been used as a chemical probe for studying the mechanism of action of other inhibitors. DMACA is an inhibitor of the enzyme discorhabdin synthase from the bacterium "Burkholderia cepacia" and didemnin synthase from the bacterium "Pseudomonas putida". DMACA binds to monolayers with a Kd of 9 μM and has been used as a fluorescent resonance energy transfer (FRET) probe for studying protein interactions.
