Dichlorophenylphosphine is a steric interaction inhibitor that binds to the active site of DNA gyrase, which is an enzyme that maintains the integrity of bacterial DNA. This binding prevents replication and transcription by preventing the unwinding of double-stranded DNA. Carbon disulphide is used in this reaction as it can react with phosphorus to form a reactive intermediate, which reacts with the phosphate group on the enzyme. This reaction is shown below:
Phosphorus + Carbon Disulphide → Phosphonium ion + Sulfur Dioxide
Phosphonium ion + Active Site → Phosphineoxide radical + Inactive Site
The phosphineoxide radical attacks the phosphate group on the enzyme, leading to its inactivation. The high values for dichlorophenylphosphine are due to its high binding constants and low expression levels.
