Vandetanib is a drug that belongs to the class of tyrosine kinase inhibitors. Vandetanib is used for the treatment of cancer, including renal cell carcinoma and medullary thyroid cancer. Vandetanib inhibits the growth of cancer cells by blocking the epidermal growth factor receptor (EGFR) and other tyrosine kinases. It also has potent inhibitory activity against other protein kinases such as c-Src, Bcr-Abl, and Abl. The pharmacokinetics and metabolism of vandetanib have been studied in healthy volunteers following single doses up to 240 mg. Vandetanib is metabolized in humans by cytochrome P450 3A4 (CYP3A4) enzymes to an active metabolite that has a longer half-life than vandetanib itself. This active metabolite is further metabolized by glucuronidation reactions or by oxidation of its n-oxide group to form
