Lomustine is a nitrosourea that inhibits the growth of malignant cells by causing DNA damage. It binds to the tumour suppressor gene and has been shown to be effective in the treatment of leukemias and brain cancer. Lomustine can be used as an adjuvant therapy for patients with resected brain tumours, where it has been shown to decrease the incidence of post-operative recurrence. Lomustine also acts as a mutagen, which may account for its ability to induce malignant transformation in mammalian cells.
