Dimethyl N,N-diethylphosphoramidite is a nucleophilic reagent that is used in peptide synthesis. It reacts with benzyl esters to form tripeptides in high yield. The reaction proceeds through nucleophilic attack of the phosphite on the carbonyl group of the benzyl ester, followed by protonation and elimination of diethyl phosphate to give the tripeptide product. Dimethyl N,N-diethylphosphoramidite can also be used for hydrogenolysis. A number of halides react with dimethyl N,N-diethylphosphoramidite to form a halogenated amine in situ. This reaction provides an alternative to chlorination or bromination reactions and is applicable for the synthesis of peptides containing hydroxy groups or electrophiles such as chlorine, bromine, and iodine.
