Description

Atorvastatin is a drug used to lower cholesterol levels in the blood. It belongs to a class of drugs called HMG-CoA reductase inhibitors, and it works by blocking an enzyme that plays a role in the production of cholesterol. Atorvastatin calcium is a salt form of atorvastatin that is absorbed better than the free acid form. The sequential synthesis of atorvastatin lactone and its hydrolysis yields atorvastatin calcium. This drug can be synthesized from tert-butyl, silver nitrate and atorvastatin lactone, which are reacted together with hydrogen peroxide under acidic conditions to produce the desired product. Silver nitrate is then added to convert any unreacted starting material into silver, which is removed by filtration. The final step in this process involves reacting the product with triflate or organic chemistry to yield atorvastatin calcium.
