Diethyl acetylmalonate is a bromodomain inhibitor that can be used as a cancer therapeutic. It binds to the bromodomains of the histone protein, inhibiting transcription and replication of the cell's DNA. Diethyl acetylmalonate also inhibits the enzyme acetate extraction in cells, which leads to an accumulation of malonyl-CoA and inhibition of fatty acid synthesis. This drug is activated by Friedel-Crafts reactions with quinoline derivatives and hydroxyl groups, which produces reaction products that are also active methylene compounds. The molecule is hydrolyzed by hydrochloric acid in the stomach to form an active methylene compound that can cross into cells and inhibit human immunodeficiency virus (HIV) infection.
