2,6-Dichloro-4-methylnicotinonitrile is a synthetic compound that has been shown to be an effective inhibitor of HIV reverse transcriptase. It was the first synthetic nucleoside analog to be used in the treatment of HIV infection and has been used in combination with other drugs such as nevirapine and zidovudine. 2,6-Dichloro-4-methylnicotinonitrile binds to the active site of HIV reverse transcriptase and inhibits its activity by binding to a catalytic site that contains a chloride ion. This binding prevents the formation of covalent bonds between deoxyribose phosphate (phosphate) and deoxyadenosine triphosphate (ATP). The nucleophilic attack by the chlorine atom on the phosphorus oxychloride leads to cleavage of the phosphorus oxychloride bond, which results in inactivation of this enzyme. This inhibition prevents viral replication.
