Dabcyl acid 4-dimethylaminobenzene-4â-carboxylic acid is a dark quencher chromophore that absorbs in the visible region and serves as an acceptor in FRET applications. It is widely used to measure protease activity by labelling a suitable peptide substrate with a fluorophore donor and a dabcyl dark quencher as an acceptor. The fluorophore-quencher energy transfer is related to distance (normally within 10nm). The peptide is cleaved by the protease action and the dabcyl quencher diffuses away from the fluorophore, and the fluorescence is measured. In RT-PCR, it is used at the non-fluorescent 3â terminal of oligonucleotides as a quencher.
