4,5-Diamino-2-fluorobenzonitrile (DAFB) is a highly selective radioligand for the dopamine D4 receptor. It has a high affinity for this receptor and binds with a Kd of 1 nM. DAFB has been used in vivo imaging studies and autoradiographic studies to examine the distribution of the D4 receptor in the brain. Low concentrations of DAFB have shown high affinity for the D4 receptor, but as concentrations increase, binding becomes more selective. The low affinity for other receptors is due to its lack of lipophilicity and hydrogen bond donor sites. Radiosynthesis of DAFB has been achieved through an oxidative coupling reaction using nitrobenzene and 2-chloroethanol.
