2,4-Difluoro-5-methylbenzonitrile (2,4-DMN) is a prodrug that is metabolized in vivo to its active form, 2,5-difluorobenzoic acid. 2,4-DMN inhibits the activity of the NS5B polymerase enzyme in the replication of human immunodeficiency virus type 1 (HIV-1) by competitive inhibition and prevents viral RNA synthesis. This compound has been shown to be orally bioavailable and efficacious against HIV-1 in monkeys. 2,4-DMN also inhibits the activity of other types of viruses with a nonpolar active site such as herpes simplex virus type 1 and varicella zoster virus.
2,4-DMN has been found to have little or no inhibitory effects on mammalian DNA polymerases α or β when used at concentrations up to 10 μM.
