Endothelin (ET) is a potent vasoactive peptide that is produced by endothelial cells in response to injury or inflammation. ET binds to the endothelin receptors and activates phospholipase A2, which releases arachidonic acid from membrane phospholipids. This leads to the production of prostaglandins and thromboxanes, which increase vascular permeability and induce vasoconstriction. Endothelin also causes smooth muscle contraction and relaxation in many tissues including the mesenteric, skeletal, and cardiac muscles. The binding of ET to its receptor increases blood pressure by causing vasoconstriction. ET is used as a tool for studying receptor binding in guinea pigs because it has been shown that it has biphasic effects on tracheal contractility at low doses (i.e., it causes relaxation) but at high doses (i.e., it causes constriction).
