2-Ethoxynicotinonitrile (2EN) is a drug that has been shown to have significant in vitro cytotoxic activity against cultured human MCF-7 cells. 2EN inhibits the DNA synthesis process by interrupting the metabolic pathway of the enzyme hydroxylamine, which is involved in the conversion of propenone to phenylhydrazine. It also has inhibitory effects on chloride ion channels and drug doxorubicin uptake. The cytotoxicity of 2EN is increased when irradiated with nitrous oxide gas or when mixed with other drugs such as thiosemicarbazide or thiourea. In addition, 2EN showed inhibitory effects on mitosis and cell division when combined with phenyl isothiocyanate or phenylhydrazine.
