Ethyl 4-hydroxynicotinate is a conformationally restricted analog of nicotinic acid. It has a planar conformation with two hydrogen-bonded OH groups and one methylene group in the axial position. The molecule has an ester linkage between the alcohol and carboxylate, which is hydrolyzed to release ethanol and formaldehyde. The hydrolysis reaction is catalyzed by esterases and proceeds via a chloride ion as the leaving group.
Ethyl 4-hydroxynicotinate can be synthesized by reacting ethyl chloroformate with ethanol in the presence of sodium hydroxide or potassium hydroxide. This reaction produces ethyl 4-chloronicotinate, which can be converted to ethyl 4-hydroxynicotinate by heating it with an excess of ethanol followed by treatment with hydrochloric acid.
