Etodolac is a non-steroidal anti-inflammatory drug (NSAID) that inhibits prostaglandin synthesis. It also has a pro-apoptotic effect on cells and can induce apoptosis in primary cells. Etodolac is used to treat pain and inflammation associated with osteoarthritis and rheumatoid arthritis. The mechanism of action of etodolac is not fully understood, but it has been shown to inhibit the production of pro-inflammatory substances such as β-catenin and nitric oxide, which may be due to its ability to form an intermolecular hydrogen bond between the hydroxyl group on the drug molecule and the COX-2 inhibitory site on these enzymes.
Etodolac also binds to erythrocytes, resulting in a decreased blood flow rate in rats with induced inflammation. Etodolac has been shown to have an activity index of 150 compared with 10 for ibuprofen
