2-Ethoxy-8-methoxyQuinoline is a macrocyclic inhibitor that is synthesized from 2,6-dichloroquinoline and ethylene. It binds to the active site of the enzyme adenosine deaminase (ADA) and inhibits its function by a nucleophilic attack on the chloride ion. This reaction takes place in two steps: first, the chloride ion is displaced by an electron-deficient nucleophile such as benzonitrile or methoxide to form a reactive intermediate; second, this intermediate reacts with ethylene to form the product. The nature of this reaction time depends on the solvent used. In water, the reaction time is increased because water molecules activate benzonitrile and increase its nucleophilic attack. 2-Ethoxy-8-methoxyQuinoline has been shown to inhibit cancer cell growth in primary culture and yields high melting points when it crystallizes.
