1,3,4-Oxadiazole-2(3H)-thione is a conformationally restricted analogue of oxazolidinone that exhibits potent analgesic activity. This compound contains a reactive hydrogen atom and an organic group in the para position to this atom. The presence of these functional groups results in hydrogen bonding with the benzene ring, which increases its stability and thus promotes its biological activity. 1,3,4-Oxadiazole-2(3H)-thione has been shown to have antipyretic activity against prostaglandin synthesis by inhibiting cyclooxygenase (COX) enzymes. It also inhibits COX1 and COX2 in mice at doses below those that cause significant adverse effects on body weight gain or food consumption.
