5-Formylimidazole-4-carboxylic acid methylester (5FICM) is a small molecule that has been shown to inhibit the growth of cells by blocking the TNF-α receptor. The molecular modeling data and crystallographic data indicate that 5FICM can bind to the kinase domain of TNF-α receptors, preventing activation of downstream signaling pathways. In knockout mice, 5FICM was found to reduce skin inflammation caused by LPS. It also inhibited tumor necrosis factor (TNF)-α production in epidermal growth factor (EGF)-treated human keratinocytes and inhibited EGF-induced epidermal growth. These data suggest that 5FICM may be a potential therapeutic agent for inflammatory skin diseases, such as psoriasis and atopic dermatitis.
