5-Formylcytosine (5FCC) is a cytotoxic drug that is used for the treatment of cancer. 5FCC can be synthesized from uracil, which is a common nucleobase in DNA and RNA. 5FCC inhibits transcription by binding to the polymerase chain reaction (PCR), preventing the synthesis of DNA. It also induces glycosylases, which are enzymes that remove sugar residues from proteins. The removal of sugar residues causes misfolding and aggregation of proteins, leading to cell death. This drug has been shown to inhibit the growth of skin cancer cells in mice by photochemically inducing 5FCC within the cell.
5FCC binds to dna duplexes through hydrogen bonding, which stabilizes its structure and prevents it from breaking down into uracil. This inhibition prevents the repair mechanism that normally corrects errors in DNA replication.
