Fmoc-N-methyl-D-valine is a reagent that can be used for coupling amino acids to the amino group of an N-substituted benzyl halide. This chemical is prepared by reacting the desired amino acid with collidine in the presence of triphosgene and n-alkyl bromoacetate, followed by cyclization under sterically demanding conditions. The efficiency of this coupling reaction has been improved by using solid phase condensation on a polystyrene resin with an immobilized carboxylic acid group. Solid phase synthesis enables the efficient preparation of large quantities of Fmoc-N-methyl-D-valine with high purity and yields.
