Febrifugine is a coumarin derivative that has been shown to be active against malaria in vitro. It is an experimental model for the study of autoimmune diseases, such as multiple sclerosis. Febrifugine also has antimicrobial activity and can be used to treat infectious diseases. The drug inhibits the synthesis of proteins essential for bacterial growth by inhibiting the chorismate synthetase enzyme, which catalyzes the conversion of chorismate into isopentenyl pyrophosphate (IPP). This blockage leads to a loss of mitochondrial membrane potential and inhibition of protein synthesis. Febrifugine also exhibits pharmacokinetic properties that are similar to other anti-malarial drugs, including chloroquine and quinine.
