3'-Fluorothymidine-5'-triphosphate is a nucleotide analog that is a potent inhibitor of HIV and hepatitis B virus. It binds to the polymerase enzyme, preventing the synthesis of viral DNA. 3'-Fluorothymidine-5'-triphosphate has been shown to be toxic for human cells in culture. This toxicity is due to its ability to inhibit cellular protein synthesis by binding to ribonucleotide reductase, an enzyme required for DNA synthesis. 3'-Fluorothymidine-5'-triphosphate also inhibits transcription and replication of human mitochondrial DNA by binding to the transcription factor TFIIIA, which regulates RNA polymerase I activity.
