Hippuryl-His-Leu-OH is a peptide that inhibits the activity of angiotensin converting enzyme (ACE) at a concentration of 50 μM. It has been shown to inhibit cyclase and other enzymes in vitro. The binding of this compound to ACE prevents the conversion of angiotensin I to angiotensin II, which is a potent vasoconstrictor. Hippuryl-His-Leu-OH also has inhibitory properties against atrial natriuretic peptide (ANP), and can be used as an antihypertensive agent. This drug has been shown to have growth factor β1 activities, and can be used for the treatment of cardiac diseases such as myocardial infarction. Structural analysis shows that this drug binds to the active site of ACE, inhibiting its activity by blocking access to substrate or by altering substrate specificity.
