Nipecotic acid is a n-propyl derivative of the amino acid phenylalanine. The compound is an alkylating agent that reacts with nucleophilic sites in proteins, DNA, and other macromolecules to form covalent bonds. Nipecotic acid has been shown to have antiplatelet effects by inhibiting platelet aggregation and adenosine uptake. It also inhibits the synthesis of arachidonic acid and thus prevents platelet activation. Nipecotic acid can be used as a marker for human blood platelets due to its presence in human plasma at concentrations higher than those found in rat plasma. Nipecotic acid has been shown to inhibit platelet function by binding to the catalytic site of phospholipase A2 and blocking its activity. This prevents the hydrolysis of phospholipids into arachidonic acid and lysophosphatidic acid, which are involved in platelet aggregation.
