4-Hydroxy-1H-indole-2-carboxylic acid (4HIC) is a potent inhibitor of 3CLpro, which is an enzyme that catalyzes the conversion of proline to glutamate and pyroglutamate. 4HIC inhibits the binding of proline to the active site of 3CLpro. This inhibitor has been shown to have affinity for both 2,3-diaminoproline and L-prolylglycine. The modifications in the structure and moieties of 4HIC are responsible for its inhibitory activity against 3CLpro. Titration calorimetry experiments show that 4HIC has a potent inhibitory activity with an IC50 value of 0.5 μM in the nanomolar range. The inhibitory potency was determined by titration calorimetry experiments with an IC50 value of 0.5 μM in the micromolar range.
