2-O-Hexadecyl glycerol is an intermediate in the synthesis of phospholipids, which are important components of cell membranes. It is also a precursor for the biosynthesis of monoglycerides and diacylglycerols. 2-O-Hexadecyl glycerol is commonly found in adipose tissue and has been shown to inhibit tumor growth. This lipid can be synthesized from cholesterol by an acyltransferase enzyme that uses a fatty acid as an acyl donor. 2-O-Hexadecyl glycerol inhibits cholesterol acyltransferase, as well as other enzymes, such as protein synthesis enzymes. The steric interactions between this lipid and other lipids may also affect enzyme activities.
