Para-hydroxyatorvastatin calcium is a high-performance liquid chromatography (HPLC) analytical standard of atorvastatin. It is a prodrug that is metabolized in the liver to its active form, atorvastatin. The purpose of this study was to validate and characterize an HPLC method for the simultaneous quantification of para-hydroxyatorvastatin calcium and atorvastatin in human plasma. The validation parameters were linearity, precision, accuracy, selectivity, extraction efficiency, stability over time and bioequivalence. The validation results indicated that the calibration curves for para-hydroxyatorvastatin calcium and atorvastatin were linear over the concentration range 0.5-50 ng/mL with correlation coefficients greater than 0.9997.
