Haloperidol N-oxide is a pyridinium salt that inhibits the uptake of dopamine. It has been shown to inhibit the binding of [3H]haloperidol, a dopamine receptor antagonist, to rat striatal membranes and to inhibit the binding of [3H]spiperone, a dopamine receptor agonist, to rat striatal membranes. This inhibition may be due to competition for the uptake sites on the membrane or may be due to an effect on the kinetic properties of these receptors. Haloperidol N-oxide is also a potent inhibitor of chromatographic methods used for analysis and purification of haloperidol. This compound is synthesized by reacting haloperidol with hydrochloric acid in an acidic medium. The n-oxide group is then introduced into the molecule by oxidation with nitric acid in an alkaline medium.
